Extraction, Identification and characterization of an alkaloid Palmitine
from the calli cultures of Coscinium fenestratum L

ABSTRACT
Present study describes in detail extraction and characterization of alkaloid palmitine from the calli cultures of an endangered medicinal plant Coscinium fenestratum. Petiole derived calli cultures were established in MS medium supplemented with 2,4-D and BAP. Air dried calli cultures were homogenized with mortar and pestle then pure methanol was used repeatedly to extract until all the brown color had been removed from the material. Methanol was evaporated and the concentrated extract was separated by preparative TLC by using chloroform : methanol (8.5:1.5). The crude extract was loaded into a silica column and eluted with different fractions of solvents from increasing polarity. The yellow fluorescent fraction showed the presence of palmitine by TLC, was collected and evaporated by dryness. Yellow colored powder thus obtained was identified by comparing Rf values on TLC. High resolution mass analysis of the molecular ion, M +, 352.1544 of compound shiny yellow solid, mp 196-198 gave the elemental composition C21H22O4 (calc.352.1548) The IR and 13 C NMR spectra did not show either carbonyl or hydroxyl functional group indicated the absence of theses groups in the structure. The 1H NMR spectrum recorded in DMSO-d6 accounted for all the 22 protons in its molecular formula.
